Microglia play a crucial role in the neuroinflammatory processes underlying a wide array of brain pathologies and are increasingly becoming targeted by drug discovery efforts.
The Neuroinflammation Tissue Clearing Kit enables researchers to visualize microglial activation and distribution throughout the entire CNS by measuring Iba1 (Ionized calcium-binding adapter molecule 1).
Iba1 is upregulated during microglial activation, making it a widely used indicator of neuroinflammatory responses. Iba1 staining can be used in various CNS tissues, including the mouse brain, spinal cord, and eye.
In the figure below, the Neuroinflammation Tissue Clearing Kit was used to measure the expression of Iba1 in response to either lipopolysaccharide (LPS) or murine coronavirus (MuCoV) treatments. The zoomed in regions demonstrate that the response to LPS is relatively homogeneous throughout the brain, whereas MuCov results in a very heterogeneous patterns with the Iba1 expression level differing dramatically across brain regions. This kit allows users to measure neuroinflammatory responses like those seen below.
The kit includes validated Anti-Iba1 Primary Antibodies and Iba1 Secondary Antibodies (647), providing a complete clearing and staining solution for Iba1 visualization.
The Neuroinflammation Tissue Clearing Kit can also be used in conjunction with the users antibody of choice in the 568 channel. This allows for the simultaneous visualization of neuroinflammation and another epitope of interest. This provides flexibility in experimental design, allowing for a customized staining protocol tailored to your research question.
