Whole-Organ Tissue Clearing

The Liver Bile Duct and Other Tubular Structures Quantification service utilizes Translucence Biosystems’ modified iDISCO+ tissue clearing protocol to render intact samples optically transparent. Staining for Cytokeratin 7 (CK7), found in liver bile ducts and Smooth Muscle Actin (SMA), a marker of vascular smooth muscle cells, facilitates the visualization of the bile ducts and vasculature across the intact liver.

Light Sheet Imaging

Cleared, labeled samples are imaged on the Lightsheet Z.1 equipped with the Mesoscale Imaging System™. The Mesoscale Imaging System™ is optimized for imaging large intact tissues with unprecedented speed and resolution and is compatible with the high refractive index solutions used in tissue clearing protocols, such as iDISCO+.

Translucence’s imaging pipelines capture cellular-resolution, whole-sample images of liver bile ducts and other tubular structures, providing comprehensive insights into morphological changes induced by genetic or pharmacological manipulations.

Light Sheet Image Stitching

We employ our in-house developed software solution, Stitchy, for automated light sheet image stitching. Stitchy is a robust software solution that stitches large imaging datasets 2.5-5.5 times faster than leading commercial software solutions. Stitchy enables seamless integration with downstream image analysis pipelines by eliminating intermediate file formats.

Liver Vasculature

Visualization of liver vasculature using Smooth Muscle Actin (SMA) in the whole mouse liver. Tissue clearing and staining for SMA enables the use of lightsheet imaging to visualize the vascular networks in the liver. This approach provides robust insights into vascular networks, often implicated in liver diseases. Tissue clearing provides more comprehensive insights compared to traditional 2D slice immunohistochemistry (IHC) methods. Sample preparation was performed using the Translucence Universal Tissue Clearing Kit. This NIH-funded kit is suitable for clearing various tissue types, including mouse brain, lung, liver, spinal cord, and more. Imaging was performed on the ZEISS Lightsheet Z.1 equipped with the Mesoscale Imaging System™.
