Adapts the ZEISS Lightsheet Z.1 microscope, designed for imaging small live organisms, to image large intact tissues with unprecedented speed and resolution
Translucence has developed the Mesoscale Imaging System, which makes the ZEISS Lightsheet Z.1 microscope compatible with the high refractive index solutions used in iDISCO+ and other tissue clearing methods.
The system consists of the Imaging Chamber, a Specimen Holder and a Safety Collar/Objective Adapter. Together, these components exploit the superior optics of the ZEISS Lightsheet Z.1 microscope and tune them for cleared tissue.
The improved optical resolution allows for wide-field low-magnification objectives to be used to achieve high quality images in a fraction of the time that it would normally take. For instance, using a 2.5X objective, an entire mouse brain can be imaged with a pixel size of 1.8 X 1.8 X 12 µm in ~40 minutes.
The imaging chamber is precision-machined in San Francisco from high quality stainless steel.
The chamber is designed to increase the inner diameter and has an optimized shape to accommodate large tissues such as mouse brains.
The volume of the chamber is 42% greater than the standard Zeiss chamber.
The design of the chamber windows prevents leaking of the immersion solution and allows for easy replacement of glass when needed.
The side windows are positioned for light sheet optimization, centering beam waists at the detection optic’s focal plane.
A positioning mechanism facilitates adjustment of the imaging objective focal plane according to the refractive index of the high RI solution used.
The Translucence Specimen Holder consists of 3 components: the holder arm (1) hangs from the Z.1 microscope above the chamber and is assembled with a clamp (2) that was designed to hold custom-designed tissue mounts (3).
The system comes with 5 different mounts for tissues of various sizes
We are open-sourcing the design such that end-users can design and 3D print customized mounts that are easily exchanged.
We can manufacture designs shared with us and will make all shared designs available to users.
